Salient Characteristics Azure Biosystems Sapphire Biomolecular Imager List of Salient Characteristics, unique features capabilities. System must offer optimal technology for each fluorescent dye. System must include optical modules capable of detection of near-infrared dyes (e.g., 685 and 784nm emission). System must offer expandability to additional optical modules that will enable detection of 3 or more fluorophores. System must offer expandability to additional models of imaging, such as chemiluminescence detection and phosphorimaging. Simultaneous multi-channel detection must be possible for fluorescent multiplex samples. With maximum scan speed 250 mm/s, must be able to image 3-channel fluorescence on a 6.5 x 9 cm blot at 100 mm resolution in 5.6 min. Laser lifetime must exceed 40,000 hours of operation time before failure. This keeps maintenance operational costs extremely low. Dynamic range of the system must be at least 6 logs. Laser must be integrated into a precision scanning mechanism capable of scanning at resolutions between 5 m-1000 m. Data output must be a 16-bit TIFF file for each channel of detection. The system must be able to capture images with Extended Dynamic Range (EDR) enabling 24-bit data. The system must be able to perform SmartScan scans where the optimal intensity of the lasers is determined by the system, without saturation. The system must utilize a sealed, flat bed scanning surface to accommodate varying sample types (membranes, gels, multi-well plates, small animals, and slides). The system must have a 25 x 25 cm field of view to accommodate large samples or multiple smaller samples (blots, 96 well plates). There must be a 4 cm clearance from glass platen to the inside of the scanner lid to support scanning of larger samples. The System must be enabled with connections for external anesthesia equipment. The system must support a 7 mm focal range (from 1.0 mm below glass platen to +6.0 mm above glass platen) and be able to take images at multiple focal heights with one command from the user to create a Z-stack of images. The system must have an automated channel alignment procedure so when optical modules are swapped, the alignment can be quickly performed for accurate imaging. Focusing must be adjustable 1 mm below to 6 mm above the scan bed to obtain best signal-to-noise ratio. Must be able to create up to 4 channel/dye images within the Capture Software. Must be able to independently scan more than one region of interest (ROI) with independent scan settings. Must be able to save imaging protocols to increase sample throughput. Must be able to merge images scanned at different focus settings on the Z axis to generate a single TIFF file. Must be able to create an animated .gif file of images scanned at different focus settings on the Z axis.
