The Veteran s Administration (VA) Office of Research and Development, Regional Procurement Office East in Pittsburgh, PA intends to negotiate on a sole source basis with the Fluidigm Corporation based in San Francisco, CA. The requirement is to provide one (1) Super High-Dimensional High-Resolution Imaging System for planned VA medical studies. The system required shall include the following characteristics as minimum specifications: Specs Imaging System Package for Currently installed Mass Cytometer Capability to be fully integrated with current Helios CyTOF Mass Cytometer, with the following features: Nd:YAG deep UV solid state laser for ablation at 20 Hz (tuning only) and 100/200 Hz (ablation). Perform sequential laser ablation of protein bound metal-tagged antibodies with a mass range of 75-209 Da from individual regions (<1µm2) regions of fixed tissue sections or cells smears which are then individually atomized and ionized, separated by mass and quantified. Optical system to include UV focusing components of laser source for <1µm2 ablation spot size of solid tissue, or cells on slides. Ablation chamber with a sample slide positioning stage for sub-micron positioning accuracy. Coupling system for connection between the Hyperion Tissue Imager and the Helios for transferring ablated sampled streams to the ICP for analysis. High-parameter analysis using metal-labeled antibodies with up to 52 elemental tags available without re-staining and 135 channels total. Ability to determine the elemental composition of each micron separately, with data from its (x,y) position noted and rasterized at a frequency of 200 pixels/second into a digital image, which is then analyzed by advanced software, single cell segmentation, and bioinformatics algorithms. Super high sensitivity, to include 0.3% sensitivity for 159Tb. Detection limit of 400 antibody copies per µm2. Dynamic range of 4 orders of magnitude. Automated calibration. Cross-cell contamination of <2% (selected direction). Crosstalk pixel to pixel <15% at 200 pixels/second. Wet tissue thickness for full ablation <7µm. Addressable sample size on slide >15mm x 45 mm. Optical view of sample >250µm x 250µm. File type: TXT, multipage TIFF, OME-TIFF, MCD. Scan area >1mm2 / 2 hr (at 200Hz). Tissue Imager 101 Training Training to include: 3-day intensive overview of the Hyperion Imaging System and Imaging Mass Cytometry. Hands-on experience and mastery of the Hyperion Imaging System operation and maintenance. Introduction to experimental design and data analysis. All necessary Fluidigm reagents for training Tissue Imager 201 Training Training to include: 3-day operation and maintenance refresher visit to enable higher dimensional experiments and new research projects. Dedicated time for both operators and researchers Tissue Imager Preventive Maintenance Preventive maintenance service included The requirement will take place at a VA Charleston Healthcare System located in Charleston, SC. The sole source is pursuant to FAR 6.302-1 Only One Responsible Source and No Other Supplies or Services Will Satisfy Agency Requirements.
