The Department of Veterans Affairs, VA Pittsburgh Healthcare System, requires the procurement of the Olympus automated fluorescence microscope (IX83) system. A VA Pittsburgh ongoing project requires that a large amount of imaging experiments be conducted to quantify brain lesion volume, assess changes of protein expression with immunofluorescence staining assays, and analyze morphological structural changes of ischemic brains. The key specialized performance requirements of the system are as follows: User friendly software i with an intuitive layout that allows for complete customizability between users and requires very little training. Capacity for epifluorescence imaging, whole brain imaging and stitching, and capturing z-stacks. A LED light source that can view from UV to IR, allowing any fluorophore to be detected. No lasers, warm up, or cool down period is required. A base system with 5 filter cubes (DAPI, FITC, CY3 and CY5) with a frame can accommodate a two-deck configuration, housing up to 14 filter cubes. A six-mount electronic lens revolver, equipped with 4-40x apochromatic objectives, that can house any combination of 2-100x objectives. Apochromatic objectives enable high-precision imaging free from image shift, eliminating the need to change objectives when switching between phase contrast and fluorescence. A built-in monochrome camera that can enable clear fluorescence imaging with low excitation light, preventing photobleaching and phototoxicity.
