The Chromatin and Gene Expression Group at the National Institute of Environmental Health Sciences is conducting research into the contributions of genetic and epigenetic diversity in the generation of induced pluripotent stem cells (IPSCs) from patient-derived dermal fibroblasts (DFs). The process of generating IPSCs from DFs is referred to as “reprogramming.” Reprogramming is a critical part of the expanding fields of precision and patient-specific medicine. Using our established cohort of DFs, we have identified key factors that contribute to the efficiency of the reprogramming process and result in disparity between individuals. However, the underlying genetic and epigenetic differences responsible for this disparity between individuals remains unclear. We require chromatin immunoprecipitation with sequencing (ChIP-seq) services to characterize the epigenetic profiles of our cohort of DFs. Our goal is to utilize these ChIP-seq services to determine how ancestry-, sex-, and age-related epigenetic differences contribute to differences in reprogramming efficiency.
