Base Notice: NIMH 10X Genomics Library Prep Kits Notice Type: Sources Sought Posted Date: 9/13/2022 Classification Code: 65 – Medical/dental/veterinary Equipment/supplies NAICS Code: 334516 – Analytical Laboratory Instrument Manufacturing NOTICE INFORMATION Agency/Office: National Institutes of Health Location: National Institute on Mental Health Title: NIMH 10X Genomics Library Prep Kits Description(s): The National Institute of Mental Health (NIMH) Human Genetics Branch (HGB) are requesting the library prep kits in order to prepare single-cell sequencing libraries for for 80 samples, 64 as single-cell RNA-seq and 16 as a combination of single-cell RNA-seq and ATAC-seq. The samples will be processed by the National Human Genome Research Institute (NHGRI) Microarray and transcriptomics Core. A major focus of our laboratory is to study the impact of genetic risk alleles on gene expression in postmortem human brain and in neural cells derived from induced pluripotent stem cells. Toward this end, we have previously spent several years studying the effect of psychiatric risk allele in bulk RNA sequencing. However, the insights we could obtain in bulk tissue are limited by its inherent heterogeneity and averaging across variable mixtures of cell types. These severe shortcomings of bulk RNA sequencing can now be overcome using single-cell RNA-sequencing. By sequencing individual cell populations in genetic risk allele carriers, we will be able to resolve cell types and study the effect of genetic risk alleles on specific cell populations in post-mortem human brain and different stages of neuronal differentiation in our iPSC-derived models and brain samples from animal models. To study potential transcriptional mechanisms underlying the gene expression changes, we will use a combination of gene expression and open chromatin profiling for the profiling of single cells in the postmortem brain samples.
