The Section on Medicinal Chemistry (SMC) seeks contractor support for chemical reagent/assay development and laboratory testing services to produce grouped ADME assays and perform a pharmacokinetic study in C57BL6 mice. The work supports evaluation of novel molecular entities for metabolic and alcohol-associated diseases by using cell-based in vitro testing and mouse pharmacokinetic studies to assess oral availability. A Caco-2 permeability assay will be included to predict oral absorption by modeling human intestinal epithelium and evaluating passive diffusion, active transport, and efflux processes such as P-glycoprotein. The resulting data will help inform structure-activity relationship optimization, improve compound permeability, and reduce transporter-related liabilities that could hinder bioavailability.
