Background and Objectives: Understanding the molecular signature of human brain is essential to study pathogenesis of neurodegenerative diseases. The high protein dynamic range of brain tissue hinders the protein identification and quantification using traditional mass spectrometry-based proteomics. Using a high-throughput protein biomarker discovery platform to study the proteomics in post-mortem frontal cortex tissue novel biomarkers will be investigated. This platform uses partially complementary oligonucleotides to the bait proteins and measured by Next Generation Sequencing (NGS). Our goal is to identify potential therapeutical targets and biomarkers in patients with early-onsite and late-onsite Alzheimer’s disease. Project requirements: Independently and not as an agent of the Government, the Contractor shall furnish all the necessary services, qualified personnel, material, equipment, and facilities, not otherwise provided by the Government as needed for: Contractor shall perform protein biomarker discovery services, Proximity Extension Assay (PEA), on 50 samples of post-mortem frontal cortex tissues provided by the Government. Contractor shall utilize a PEA protein panel of no less than 5400 target sites. Contractor shall ensure PEA will utilize partially complementary oligonucleotides to the bait proteins and measured by Next Generation Sequencing (NGS) NovaSeq 6000 platform. Contractor shall return frozen, unused samples to Government within 2 weeks after completion of services. Contractor shall provide R scripts for data analysis. And shall send data to Government via cloud server. Contractor shall provide support and assistance with data troubleshooting when required by the Government.
