This notice is strictly for research and information purposes only. Background: Considerable research interest has been focused on the possible role of bioenergetic derangement in the progress of AD. In particular, it is unclear to what extent bioenergetic deficits in the brain, with its high energy demand, contribute to the development of AD or are consequences of pre-existing AD pathology. Moreover, the connection between these deficits and other pathological processes observed in AD, such as vascular dysfunction, needs elucidation. Fortunately, 31P MRS provides a completely non-invasive means of quantifying high-energy metabolites such as phosphocreatine (PCr), inorganic phosphate (Pi), nucleotide phosphates such as ATP and NAD/NADH and phospholipids such as phosphocholine and phosphoethanolamine. 31P MRS directly detects the 31P nucleus, which is non-radioactive and has a natural abundance of 100% and therefore requires no isotopic labeling or enrichment for observation. Like other magnetic resonance techniques, 31P MRS is spatially-selective, offering localized bioenergetic data for specific regions of the brain, and can be applied longitudinally. In addition to measuring the concentrations of phosphorus-containing metabolites in living tissue, 31P MRS can be used to calculate intracellular pH from the chemical shift difference between the 31P resonances of Pi and a reference peak such as that of PCr. Further, techniques such as saturation transfer and chemical exchange-progressive saturation (CUPS) can be applied to measure the rate of and flux through the creatine kinase reaction, which generates ATP from PCr in response to increased energetic demand. Thus, 31P MRS is an ideal means of probing metabolic changes in the brain that accompany the onset and progression of AD. Purpose and Objectives: The purpose of this acquisition is to procure a phosphorus-proton (31P/1H) array head coil for phosphorus brain imaging on NIA's 3T Philips MRI scanner Description: Contractor will provide the following: 31P / 1H head coil array; part number P-XQS-X08LE-HQ-030 Salient characteristics: Must be capable of transmitting and receiving at the phosphorus-31 (31P) frequency of a 3T MRI scanner (51.7 MHz) to acquire a phosphorus-31 spectrum of a selected brain region. Must be capable of transmitting and receiving at the proton frequency of a 3T MRI scanner (127.7 MHz) to generate proton images for phosphorus voxel placement and registration. Must fit within the body coil of a Philips 3T Elition X / MR7700 MRI scanner and fasten securely to the scanner bed to avoid image artifacts. Must have an inner diameter of at least 26 cm to comfortably fit subject’s head. Must have an active coil length of at least 24 cm to scan the entire brain in the superior-inferior direction. Must have a radio-frequency and DC electrical interface compatible with a Philips 3T Elition X / MR7700 MRI scanner (via Philips-supplied adapter box if needed). Must include probe description files and any other code needed to interface the coil to the scanner’s acquisition software (Gyroscan R5.9 / R11.1 or above). Must incorporate quadrature polarization on proton channel to maximize efficiency for 1H spin excitation and signal detection. Must be capable of exciting and receiving a proton NMR spectrum of a standard 85% phosphoric acid phantom with a minimum signal-to-noise ratio of 20:1 in one scan and a maximum line width (full width at half maximum) of 10 Hz in order to produce proton images of sufficient quality to allow reliable phosphorus voxel placement. Must incorporate quadrature polarization and active decoupling on phosphorus-31 transmit channel to maximize efficiency for 31P spin excitation. Must demonstrate ability to excite phosphorus-31 (31P) nuclei in a standard 85% phosphoric acid phantom with flip angles from 10 degrees to 270 degrees with an error of no more than 5% from the theoretical sinusoidal dependence of integrated signal on flip angle. This is needed to prevent artifacts in spin-echo and stimulated-echo experiments in which 31P must be excited with precise flip angles. Must incorporate eight phosphorus-31 receive channels and active decoupling to maximize sensitivity for 31P signal reception. Must receive a phosphorus-31 signal from a standard 85% phosphoric acid phantom with a minimum signal-to-noise ratio of 20:1 in one scan and a maximum line width (full width at half maximum) of 10 Hz in order to achieve adequate sensitivity in human brain spectroscopy experiments.
