The Genetics & Biochemistry Branch (GBB) ongoing research at the NIDDK involves work that requires cryosectioning of frozen sections of difficult tissues which will be examined by the confocal microscope. The Leica CM 1950 instrument is essential for ongoing projects involving the establishment of reporter mouse that records the activation of G protein receptor signaling with the expression of green fluorescent protein. In these studies, the mouse tissues are sectioned and examined by the confocal microscope to identify cells that express the green fluorescent protein. The system achieves frozen sections of paraffin-quality as thin as two microns. The sections are wrinkle-free, uncompressed, fully intact and bonded to the microscope slide. The system offers many advantages over conventional methods, including speed, high precision and predictability, and reduced tissue wastage. The system uses no additional consumables and requires minimal maintenance. Leica Microsystems is the only vendor with a complete replacement parts inventory, and the sole vendor that provides the necessary services related for maintenance and repair of the instrumentation. Only Leica Microsystems personnel possess the requisite authorized qualification(s), product knowledge and specialized training to provide such services.
