The USDA-ARS-SWRU in St Paul, MN is in need of a Fluorometer required for measurement of chromophoric dissolved organic matter (CDOM) samples from environmental water samples. This instrument is being procured to measure dissolved photo reactive matter in solution (mostly environmental water samples from streams, rivers, and lakes). Some of these compounds will be strongly fluorescing while others weakly. Samples will include a mixture of compounds at widely different concentrations. The instrument must therefore be able to measure complete fluorescence Absorption Transmission Excitation?Emission Matrices (ATEEMs) to allow the constituent compounds and their individual concentrations to be determined. Because it is anticipated that many such samples will need to be measured, it is mandatory that the instrument be able to rapidly complete such measurements. It is also vital that it can measure a wide range of concentrations without requiring dilution. The instrument software must automatically standardize the data for comparison with data from other labs and instruments. The accuracy of spectra produced must be verifiable by measuring the NIST fluorescence standard materials or DI water Raman spectra. The software must also automatically export data into a multicomponent analysis software package that will allow the creation of or utilization of existing models to identify the various compounds and calculate their respective concentrations. Finally, the instrument must be compact and rugged for potential field deployment. To measure the lowest concentration samples, the instrument must have: ? Sensitivity measured using the water Raman RMS method S/N of greater than 20,000:1. ? A subtractive double monochromator on the excitation side for greater than 10?7 stray light rejection. ? Include a back?illuminated, low noise CCD multichannel detector with >70% quantum efficiency to allow simultaneous measurement of all emission wavelengths to reach the highest possible S/N. ? Include a cooled detector for minimal increase in noise for longer measurements of extremely dilute samples. To measure the highest concentration samples, the instrument must be able to: ? Simultaneously measure both the fluorescence and absorbance spectra from the same sample at the same time. This is essential to be able to correct for inner?filter effects of a concentrated sample. ? Have greater than +/? .01AU absorbance accuracy at up to 2 AU. To meet sample throughput measurement requirements, the instrument must be able to: ? Measure a complete EEM over the entire 230?800 nm excitation and emission range of interest in less than 1 minute. ? Simultaneously measure the sample absorbance without requiring transfer to another instrument. ? Include a multichannel detector to allow simultaneous measurement of all emission wavelengths for fastest possible acquisition at a given S/N. ? Compile all data channels automatically to present ATEEM data. ? Allow integration and use with a multiple sample auto?sampler To guarantee that measured data is independent of instrumental influence, the instrument must be able to: ? Automatically produce wavelength and intensity corrected spectra. ? Include a spectrally?corrected CW xenon lamp for excitation to guarantee reproducible spectra across the entire measurement range of 230?800 nm while minimizing fluctuations and drift. ? Include a reference channel to normalize any remaining source fluctuations, drift. To format the collected data, the instrument software must: ? Include single?button routines to automatically correct data for inner filter effects, Raleigh line scattering, and water Raman scattering. ? Be able to automatically normalize the intensity scale in either Quinine Sulfate or Raman units ? Seamlessly transfer data for analysis to a multivariate modeling and analysis package that uses the industry standard Parallel Factor Analysis (PARAFAC) method as well as other modeling options.
