Statement of work The United States Department of Agriculture, Agricultural Research Service, Plant Science Research Unit, Raleigh, North Carolina requires: Acquisition of laboratory consumables for extracting DNA from large populations of plants to facilitate screening of the plants using marker and genomic assisted selection and sequencing. The LGC oKtopure automated DNA extraction instrument is used to isolate 800-1600 DNA samples/day from plat tissue for high-throughput screening using PCR based markers to provide data to plant breeders who have submitted the samples. These reagents, tips and 96-well magnet plates are key to maintaining sample integrity without risk of contamination and to meet instrument requirements. Requirements: 1. Customized sbeadex magnetic bead based plant DNA extraction kits specifically designed for use on the LGC oKtopure automated liquid hadling instrument and optimized for the ERSGGL (Eastern Regional Small Grains Genotyping Lab) wheat & barley extraction requirements. 2. AMP Elution Buffer base reagent for DNA concentration quantitation with PicoGreen 3. 200ul robotic pipetting tips that meet the specified requirements of the instrument and the reagent plates. 4. 96-well storage plates are calibrated to the magnets on the instrument and the reagent volumes required for washing the DNA samples. 5.
