This is NOT a request for quote- USDA-ARS, Pullman, WA intends to award a sole source purchase order to University of California, Berkeley for whole genome sequencing of 105 lentil genotypes using DNAseq. The whole genome sequencing is required for these L05 lentil genotypes (USDA accessions) in order to identify the specific genes conferring high levels of genetic resistance to Fusarium root rot in lens culinoris. Lentil has a relatively large genome (4.2 Gb) so high-density coverage (10X) is necessary. University of California, Berkeley has the equipment and expertise necessary to generate high quality sequencing libraries necessary to generate high density sequence data using the most recent high through-put sequencing instrumentation. lllumina 1.0 Gb NovaSeq 54 150 PE generates 37 million reads for a total of 42OO Gb of the highest quality sequence data (quality score +Q30). To summarize, this data will allow us to identify the specific genes conferring genetic resistance to Fusarium root rot. The gene identification will allow the plant breeders to efficiently identify new lentil cultivars with genetic resistance to Fusarium root rot. We have completed skim sequencing of lentil accessions at the University of California, Berkeley that has been published and we require comparable data to continue our research. This is for joint research through a grant. This was customized sequencing through the University of California and was used for publications. This vendor is required for consistency in already completed research and use of another vendor even if using the same specifications can have different methods or results due to lab conditions that would not be comparable to past research, and not be valid for continued research. Our justification to award is in accordance with Federal Acquisition Regulation (FAR) 6.302-1 - Only one responsible source and no other supplies or services will satisfy agency requirements.
