The Division of Intramural Research (DIR), Research Technologies Branch (RTB), Research Technologies Section (RTS) Flow Cytometry Unit (FCU) & Flow Cytometry Section (FCS) (collectively “Program”) provides intramural National Institute of Allergy and Infectious Diseases (NIAID) investigators with application-specific flow cytometric technologies, including instrumentation and multi-color analysis. In addition, the FCS provides training, consultation, method development, and analysis for experiments involving flow cytometry. This mission is accomplished through the efforts of staff with extensive flow cytometry experience and with the use of cell sorters and analyzers. Major goals are to provide access to state-of-the-art technologies, to help design and run experiments, to facilitate data interpretation, and to provide results that are of consistent high quality. The Flow Cytometry Unit of the Research Technologies Branch requests the purchase of a MACSQuant Tyto Cell-sorter (“Tyto”) which is a high-speed cell analyzer that will be used for downstream applications such as single cell genomics and CRISPR technologies. The procurement of this high-performance cell analyzer will guarantee the optimal function and therefore reliable data of the flow cytometry analysis and sorting. There is a growing need at RML for sorting fragile cells and bacteria. Current sorting equipment has limitations with purity, viability and resolution. The Tyto sorter will allow investigators very pure and viable cells for downstream applications such as single cell genomics and CRISPR technologies. This equipment will be important in conducting SARS-CoV-2/COVID-19 research and studies. Reference attached Limited Source Justification.
