THIS IS A NON-COMPETITIVE (NOTICE OF INTENT) COMBINED SYNOPSIS SOLICITATION TO AWARD A CONTRACT OR PURCHASE ORDER WITHOUT PROVIDING FOR FULL OR OPEN COMPETITION (INCLUDING BRAND-NAME). The National Institute on Drug Abuse (NIDA), Office of Acquisition (OA), on behalf of the National Center for Advancing Translational Sciences (NCATS), intends to negotiate and award a purchase order without providing for full and open competition (including brand-name) to True PharmaChem Inc., 117 Autumn Ridge Rd., Morris Plains, NJ, 07950-1197. True PharmChem Inc. has previously synthesized TEPP-46 and analogs for this project. To ensure consistency in the analogs for this project, the analogs must be synthesized and provided by the same vendor who synthesized and provided prior analogs for this project. DESCRIPTION NCATS requires the synthesis of two compounds that are activators of the M2 isoform of pyruvate kinase. The contractor shall provide: CLIN 01: Two Grams of PKM2 activators and comprehensive report. This includes: A synthetic plan for the synthesis of both analogs that builds on past methods for the synthesis of TEPP-46 One gram of NCGC00319092 One gram of NCGC00344932 One comprehensive report The required characteristics are: The PKM2 activators must be synthesized using standard methods in accordance with best practices. The PKM2 activators must be synthesized using step-wise, resulting in at least one gram of each analog. The PKM2 activators and all key intermediates must be characterized using standard tests (1H NMR, 13C NMR, LCMS trace confirming purity and HRMS confirming identity). The comprehensive report must include the synthesis of each analog including all synthetic steps and characterization data (1H NMR, 13C NMR, LCMS trace confirming purity and HRMS confirming identit See the attached Request for Quotations (RFQ) and corresponding documents for complete requirement details.
