FDA Filth Labs perform many Association of Official Agricultural Chemists (AOAC) and Laboratory Information Bulletin (LIB) methods that employ autoclave digestion, such as frequently used AOAC 972.37 Light Filth in Baked Goods with Fruit and Nut Tissues and AOAC 969.41 Light Filth in Alimentary Pastes. These methods most often analyze six – 225g subsamples of product contained in 2L beakers covered with watch glasses digested in an autoclave chamber. ARL Filth Group has an autoclave, but its chamber capacity is maximum six 2L beakers, and this autoclave has track pieces in the chamber that have broken due to age and wear. A new autoclave is necessary to meet SOP-000280 requirements to process a reagent blank under the same conditions as the regulatory sample subs. This reagent blank is used to assess contamination introduced during the extraction phase and shall incorporate the same equipment, glassware, apparatus, and reagents that are used in the analytical subsamples. At times, a sample may utilize a spike and spike duplicate run along with the six regulatory subs and the reagent blank. In these cases, a Filth Lab autoclave would need a chamber with capacity to hold up to nine total 2L beakers covered with watch glasses.
